In Vitro Protocol for Sterilization and Micropropagation of potato (Solanum tuberosum L.) CV. Arizona ( مقبول للنشر )
In Vitro effect of Auxin in combination with Cytokinins on callus induction and some morphological traits of potato cultivar Arizona (Solanum tuberosum L.) were studied. Sprout were isolated from potato sterilized with ethanol and NaOCl at different time concentrations then cultured on Murashige and Skoog MS medium. Results revealed that highest survival percentage 100% with less contamination 8% and 4% (C2H5OH (10s) + 2.5% NaOCl ( 10min) and 70% C2H5OH (20s) + 2.5% NaOCl (15min) respectively. 2weeks later leave and stem segments were isolated from shoots and culture on MS medium supplement with different concentrations of auxin (2,4-D, NAA) and cytokinins (BA, Kint). Between the two explants leave segments showed better response 72% for callus induction compare to 59% for stem segments. Regarding to plant hormones, combination of 2.0 NAA + 3.0 BA mg. l-1 and 2.0 2,4-D + 2.0 NAA +2.0 Kin mg. l-1 affected positively and no significant differences were found between those treatments. Furthermore, 2.0 2,4-D + 2.0 BA mg. l-1 superior positively in giving 4.40 shoots. Explants-1.
