Antimicrobial susceptibility of Vibrio cholerae Isolated from Diarrheal Patients in Najaf Province

AuthorsZahraa Yosif Mutawek, /
JournalJournal of Chemical and Pharmaceutical Sciences
Abstract

ABSTRACT Stool samples were gathered from patients with watery diarrhea matured (about 1 for >60 years) earlier organization to antibiotic operators who were alluded to Central Health Laboratory in Najaf. This study found that 27 isolate of Vibrio cholerae 52.3% male and 47.7% female. With respect to the serotyping of V. cholerae this research obtained whole isolates possess just Inaba serotype. The virulence factors were distinguished in all isolates was deliver β-hemolysis on blood agar. The highest one appeared at the (11-20 and 21-30) years age group with 29.6%, followed by others at age groups (>60) years and (41-50) years with 11.1%. The low isolation frequency was recorded at (51-60) years in a percent 3.7%. The consequences of this test demonstrated that V. cholerae has an extraordinary resistance to most commonly antibiotics utilized in hospitals, V. cholerae indicated diverse susceptibility towards antibiotics utilized in this study. The highest rate of resistance is seen with ampicillin 100%, nalidixic acid 89% and ciftazidime 85% and moderately resistance to pipemidic acid 70%, tobramycin 67%, aztreonam 63%, cefotaxine 48%, amikacin 41% and norflaxacin 41% whereas is relatively lower resistance toward azithromycin 37%, erythromycin 33%, ceftriaxone 33%, chloromphenicol 22%, tetracycline 11% and ciprofloxacin 7.5%. The consequences of this study showed that V. cholerae isolates were obtained to be a remarkable sensitive to Imipenem (100%) as well

Abstract

Pseudomonas aeruginosa is a common bacterium in the environment. In humans, it is an opportunistic pathogen that is a prevalent cause of hospitalacquired infections. Currently, routine bacteriological culture on selective/non-selective culture media is the cornerstone of microbiological detection. The aim of this study was to compare isolation rates of P. aeruginosa by conventional culture and molecular (PCR) detection of P. aeruginosa isolated from intrauterine devices. Out of the 98 intrauterine devices collected from women in three hospitals in governorate Al-Najaf, Iraq, only 20 (30.5%) isolates belonged to P. aeruginosa, identified by diagnostic tests including cultural characters, biochemical tests and confirmed by PCR technique. The PCR method of gene algD GDP mannose was the best methods for diagnosis, which has led to isolate and diagnosis 20 isolates of P. aeruginosa from intrauterine device.

DETECTION OF MACROLIDE RESISTANCE GENES ERMB AND MEFA/E IN IRAQ

AuthorsZahraa Yosif Mutaweq, Dr. Hawraa A. Al Dahan, Dr. Habeeb S. Naher
JournalWorld Journal of Pharmaceutical Researc
Abstract

ABSTRACT The results of amplification of ermB gene by monoplex-PCR revealed that 83.8% of S. pneumoniae isolates gave positive result for detection of ermB gene, only 16.2% isolates of S. pneumoniae gave negative result for detection of ermB gene. The results showed that 100% of S. pneumoniae isolates had mefA/E gene. The presence of mefA/E gene was an absolute predictor of phenotypic. Multiplex PCR results showed that two amplification bands of 640 bp (ermB) and 346 bp (mefA/E) were observed in 83.8% of isolates showing a corresponding in the present of two bands, while only 16.2% isolates of S. pneumoniae appeared one band (mefA/E gene). All the S. pneumonia isolates revealed resistance to erythromycin antibiotic were tested for the ability to produce M phenotype by using the double-disk test. The result revealed that (18.9%) isolates showed an M phenotype, 81.1% isolates showed a constitutive MLSB phenotype, and no isolates (0%) showed an inducible MLSB phenotype. M phenotype isolates: 85.7% of M phenotype isolates harbored the mef(A/E) gene, and 14.2% harbored the mef(A/E) and erm(B) genes. MLSB phenotype isolates: 100% of MLSB phenotype isolates had the erm(B) and mef(A/E)genes.

Abstract

Abstract- This study provides genotypic data on 74 isolates of Streptococcus pneumoniae recovered from lower respiratory tract infections. In this study, some of them were detected by traditional phenotypic methods while others detected by phenotypic and then genotipically by using monoplex-PCR technique. The results showed that all (100%) isolates were encapsulated isolates. The molecular detection of cspA gene by using specific primer for S. pneumoniae revealed positive amplification for all isolates (100%). The results showed that overall, 89.2% of investigated isolates contained the lytA gene. In spite of, all (100%) of pneumococcus isolates have autolysin phenotypically. The results of this experiment indicated for positive amplification and it has been found that ply gene is found in 97.3% of pneumococcal isolates. In this study, the nanA and nanB genes were found in all S. pneumoniae isolates (100%) and (62.2%) respectively
