JournalBacterial Empire
Abstract

Background and objectives: Probiotics was redefined as “live microorganisms administered in inadequate amount that confer a health profit on the human for that the aim of this study were possibility of using probiotics extract with some supporting substances in antibacterial that resistant antibiotics. Materials and methods: The methods of this study included isolated and identification of bacteria from postoperative wound infection of 50 women subjected to surgical birth process then selected the P.aeruginosa isolates and scanning the sensitivity test of some cephalosporin type of antibiotics and determine the presence of blaOXA-1 gene, also the study had tested activity of two types of probiotics (Bacillus clausii and Saccharomyces boulardii )extract with some supporting substances (acetic acid and citric acid) as antibacterial. Results: The results of identification of bacterial isolates showed that 22(44%) isolates that related to Pseudomonas aeruginosa, 5(10%) isolates related to E.coli, 10(20%) isolates related to Klibsella spp., 10(20%) isolates related to Staphylococcus aureus and 3(6%) isolated related to Protus mirabils, and Overall, 22 isolates of P.aruginosa were analyzed by Kirby Bauer disk diffusion method and notice the resistance or sensitive to cephalexin, cefotriaxon and cefpime where the results confirmed 14 cephalosporin resistant isolates of Pseudomonas aeruginosa. Also detect blaOXA-1 gene by genotypic test. Amplification of b-lactamase genes shows the presence of blaOXA-1 (at 427 bp line) on among (10/22) study isolates. The results of antibacterial activity showed that inhibition zone of probiotics extract only (14and 16)mm to S.bouldarii and B.clausii respectively but it showed synergism effect when supporting the S.bouldarii and B.clausii extract with acetic acid where (18 and 20.3)mm respectively while (19 and 23.5)mm with citric acid respectively with significantly increasing differences when subjecting the bacteria to cefime ,ceftriaxone and cephlaxin antibiotics so,the inhibition zones were(1.3,10.3 and 0)mm respectively.

Abstract

Background and objectives: Cancer remains a global problem of health, and has been recorded as one of the causes of death after heart disease Natural products from plants, the environment and microorganisms are leveraged for the purpose of fighting cancer. Actinobacteria have been recognized as main sources of bioactive natural products as early as in the 1950s, for which about half of the secondary metabolites revealed, including enzymes, antibiotics, immunosuppressive, and anti-tumour agents Materials and methods:The methods of this study included isolated and identification of bacteria from soil samples and identified by morphology characters and biochemical test. Subjected extract of actinomycetes to HPLC purification then collected purified fractions then analyzed by GC-mass. After the fractions were mixed with liposome nanoparticals which tested activity on HT29 colon cancer cell line. Results: The results of identification of bacterial isolates showed the colonies growing on a SNA medium were morphologically identified where the colonies were well-growth and had a gray color, not producing dyes in the medium. The results of the biochemical tests indicated that isolates were amylase, catalase, and gelatinase producing isolates and non-lipase producing, non H2S production and consuming urea, while the carbon consumption test indicated the isolates' ability to consume starch, glucose and sucrose respectively. While The results of preparative HPLC revealed that 4 fractions analytical HPLC with (50 % HPLC-grade acetonitrile) at 254 nm and cycling up was employed to increase the separation efficiency. The chemical composition of the HPLC fractions using GC-MS showed the identification of many components example ( Hexadeconic acid , Octadecanoic acid, ethyl ester and Fumaric acid). The results of In vitro anti-tumor cytotoxicity showed that all four nano purified fractions were applied on HT 29 colon cancer cells and exhibited significantly differences compared with control.

Abstract

The present study was designed to evaluate efficiency of crude Pleurotus ostreatus extract as immunstimulater activity by investigate the contribution of some immunological parameters in response to crude extract in albino rats. The effective dose of crud extract were determined by treating the rats with different dose of extracts then evaluated the complete blood count (CBC). The result show that 100mg/kg dose was the most efficient dose in stimulating immunity so these dose chosen to further study. Three groups of rats (4 in each) were used, first group was immunized intraperetonialy with 3 dose per 14 day, second group immunized similarly with 6 dose per 14 while third group was gave intraperetonialy phosphate buffer saline (PBS) as control group. Blood samples were collected at 2 weeks last dose. Immunity parameters were detected by ELISA test (TLR4, IL2 and MCP1) as well as IgG, IgM, C3 and C4 by radial immune diffusion assay, in addition to evaluated Phagocytosis activity which appeared a significantly Pd”0.05 increased at 6dose per 14 days during 1 and 24 h (32.5 and 69%) compared with control (19 and 26.5%), respectively, also all tested cytokines (IL2, TLR4 and MCP1) were significantly P≤0.05 increased after 6 doses. 14 days compared with control treatment but only TLR4 significantly increased after 3dose. 14 days, also all humeral immune parameters tested (IgG, IgM and C3) significantly P≤0.05 increased at 6 dose. 14 days (3411, 71.35, 239)mg/dl respectively, but non significantly increased in C4, while non-significantly increased at 3dose. 14 days at all treatments.

Abstract

The present study is designed to evaluate the gained some compounds from Pleurotus ostreatus by extracted them and purification by using analytical and preparative HPLC technique, then tested the anticancer activity by invitro crystal violate (C.V) cytotoxicity assay on cancer and normal cell line, then chemical description about type of these compounds by GC-MS technique.Analytical and preparative HPLC carried out with 25%methanol with 1% formic acid mobile phase and description about 12 fractions with retenation time (9,10,11.5,12,12.5,13.5,14.7,15.5,17,18,19.5-21,21.8)respectivily at 254nm.In vitro crystal violate (C.V) cytotoxicity assay on SW-480 colon cancer cell line appeared the excellent inhibition with (1,5,6,7,12) fractions with (1000,500,250,125,62.5 μg/ml), while the same fractions have no effect on vero-101normal cell line.GC-MS analysis data gave us the most probability anticancer compounds in these fractions such as Quinoline, 1H indol, Silane compounds and N-hexadecanoic acid.
