Abstract

Antibiotic resistance is a significant issue that represent a global challenge. Hence, it has been urgent and crucial to find suitable interventions to control the spread of antibiotic resistant bacteria. Plants, on the other hand are considered as the main alternative to enhance or replace drug candidates. One of them in this context is the oily palm leaves methanolic extract. It has been found to exert antioxidant properties owing to its constituents (phenolic compounds) such as tannins and flavonoids. The study involved the determination of MIC and MBC along with the synergism effect induced by a range of concentrations of leaves oil. For E. coli, the MBC value was at a concentration of 100 mg/ml while highly resistant E. coli in concentration 50, 25, 12.5 and 6.25mg/ml. For salmonella the MBC value was at a concentration of 50 mg/ml while MIC at 200mg/ml. it can be concluded that the combination of leaves oils with antibiotics can be effective against resistant strains to traditional antibiotics.

Abstract

The study was performed to investigate the protective effect of different methanolic and oily extracts of leave and dry date of Phoenix dactylifera against oxidative stress induced by CCl4 on 49 Sprague-Dawley male rats weighed 175-200 g and aged 6-8 months. The animals were equally divided into 7 groups and assigned as follows: G1, administered 0.1 mL distilled water orally and considered control negative group (C-ve); G2, administered 0.1 mL/100 g BW corn oil (CrO); and G3 administered 100 mg/kg BW CCl4 orally for induction oxidative stress and considered control positive group (C+ve). The other four groups were initially administered 100 mg/kg BW CCl4 for oxidative stress induction and treated for two months as follows: G4, treated orally by 100 mg/kg BW of date methanolic extract (DME); G5, treated orally by 150 mg/kg BW of leaves methanolic extract (LME); G6, treated by 250 mg/kg BW date oily extract (DOE); while G7, treated by 250 mg/kg BW leaves oily extract (LOE). At the end of two months experiment, the animals were scarified, and their femurs removed for cytogenetic examination. results showed that only CCl4 group had significant increase (P< 0.05) in mitotic index compared to negative control and all treated groups. CCl4 group also recorded clear increasing in percentage of chromosome aberrations including diverse types in bone marrow cell compared to rat groups treated by date and leaves methanolic and oily extracts and negative control groups. It could be concluded that the treatment with different palm date and leaves extracts failed to overcome the genotoxic effect of CCl4 completely. Possibly, because CCl4 dosed for extended period (2 months) might cause extensive cell and genetic damages could be opposed antioxidants presented in the different palm extracts recording some but lesser chromosomal aberration compared to that CCl4 treated group.

Abstract

Veterinary pharmaceutical products are among the most important components of animal feed production, especially antibiotics. the main use of antibiotics in animals is to treat and prevent diseases, and to promote growth. Antibiotic use in animals can cause residues of antibiotics in foods such as meat., eggs , and milk. These residues can cause numerous side effects such as human transmission of antibiotic resistant bacteria, pathological immunological effects, allergies, parasitic mutations, nephropathy (gentamicin), hepatotoxicity, reproductive disorders, toxicity to the bone marrow (chloramphenicol), and even carcinogenic , sulfametazine and oxytetracycline. This analysis offers information important to veterinarians, livestock produces and public safety on the regulation of drug residues in animal products.

Abstract

Two experiments were performed, the first one was to investigate the main phytochemicals contents of Datura innoxia (DI) aqueous leaves extract and the second was to investigate the analgesic and anti-inflammatory impact of different doses of such extract against chemically and physically induced pain in mice. The result of experiment one indicated the presence of following phytochemical; flavonoids alkaloids, saponin, phenol, tannins and glycosides. In experiment two, investigate the analgesic and anti-inflammatory effects of DI aqueous leave extract in four treated groups each of 6 animals that consist of two DI extract treated gatherings (T1&T2) orally dosed with DI leaves aqueous extract at 100 and 200mg/Kg BW. compared with a group dosed Diclofenac (0.71mg/Kg BW)and Meloxicam(0.3mg/kg). The fourth group acts as a control one. in indicated that there were significant analgesic effects in dose dependent manner that means the extract possibly has central and peripheral effects. The high DI dose showed nearly the same analgesic effectas Diclofenac at dose 0.71mg/kg, T2 group and Diclofenac (0.71mg/Kg BW) group (6.7 ± 0.2 and 7.6 ±0.2) respectively after 30 minutes of administration and the effect of analgesia continued in both T2 and Diclofenac group (6. 9 ± 0.18 and 6.3 ± 0.3) respectively at 60 minutes after gives and effect appeared at 90 minute T2 and Diclofenac group (5. 7 ± 0.2 and 5.8 ± 0.12). while T1 showed pain reaction time of (5.8 ± 0.2 and 5.8 ± 0.12 and 4.6 ± 0.3) respectively at 30, 60, 90 minutes revealed less effective than T2 and Diclofenac group and all these three groups were in significant comparison with the control group that in pain reaction time (4.2 ± 0. 2 and 4.4 ± 0.1 and 4.3 ± 0.2) in different periods. The results appear caused a significant decreasing (P˂0.05) T1 (54 ±2.5) T2 (37± 3.2) and Diclofenac (0.71mg/Kg BW) (29 ± 1.2) comparison with the control group (70±1.5) in writhing test. In formalin test the results showed that both D. innoxia doses in early and late phase T2 (25.0±1.0,12.10±1.30) T1 (30±1.2,20.1±1.55) Meloxicam (0.3mg/kg) (24.7±1.0,11.10±1.20) and control (38.5± 2.20, 39±2.10) recorded their analgesic and anti inflammatory effects, this is probably due to the active phytochemical contents that posses both analgesic and anti-inflammatory effects.

Abstract

A study was performed to investigate the protective effect of different extracts of alcoholic and oil of leave and dry date of ( Phoenix dactylifera) against oxidative stress induced by CCL4 at a dose of 100 mg/kg given orally daily to five treated groups (CCl4 control (+ve) , alcoholic extract of leave (150 mg/kg) , alcoholic extract of dry date (100 mg/kg) , oil extract of leave and date (250 mg/kg), also two control dosed with vehicles (distal water and corn oil) were used . At the end of two months experiment blood were collected and oxidant and antioxidant parameter were studied in serum of all animal groups including Malon dialdehyte (MDA) and reduce glutathione (GSH). Furthermore ,the possibility of genomic polymorphism in glutathione-S- transference enzyme (GSTs) isomers (GSTT1 and GSTM1) were studied in all animal groups blood by using conventional PCR technique to understand their involvement in oxidative toxicity induced by CCL4 alone and with different palm date and leave extracts.` The biochemical results recorded a significant increase in serum MDA with significant decrease in reduce glutathione (GSH) only in CCL4 treated group compared to other extract treated one or control groups indicating lipid perioxidation effect induced by CCL4 and improvement effect of such effect by all used date palm extracts. Also polymorphism of GSTs isomers were recorded only in CCL4 group genomic result manifested by absence of GSTM1 isomer that indicate reduction in GSTs enzyme function and its involvement as one of the main antioxidant enzyme while the appearance of both GSTT1 and GSTM1 in genomic results of different extract treated groups were indicative of antagonistic ability of different date palm extract to oppose oxidative effect that cause improvement in oxidant / antioxidant balance nearly as that of control groups.

Abstract

Background : Platelets play an important role in the process of homeostasis, specifically in atherosclerosis formation when low molecular weight lipoprotein deposit in the wall of blood vessels and plaques formation ,this associated with many inflammatory response one of them is Platelet activation subsequent to the adhesion of platelets to the vascular wall results in the release of mediators that promote platelet aggregation. Aim : Statins as most drugs used for treatment hyperlipidemia have multiple effects including anti inflammatory effects , stabilization of plugs and inhibits platelets activation during atherosclerosis of the blood vessels.Materials and Methods : To evaluate the platelets activation there are many simple parameters can be made like platelets count , platelets distribution width, and mean platelets volume. Platelets parameter considered as marker about platelets activation state. Twenty one males rats were enrolled in our research , divided into three group , each group contain seven rats .First group considered as control group , second group considered was given simvastatin , third group was given atorvastatin. The drug dosing continued for 15 days after that blood was withdrawn to measure platelets count , platelets distribution width, and mean platelets volume. Results : There was no significant differences among control and treatment groups in regard to their effects on platelets count , platelets distribution width, and mean platelets volume (P >0.05). Conclusion : Neither simvastatin nor atorvastatin affect platelets count , platelets distribution width, and mean platelets volume.

Abstract

The aim of this study was to investigate the status of antioxidant glutathione peroxidase (GSH-Px) during oxidative stress in blood serum of rats subjected to oral methotrexate administration at a dose 10 mg/kg B.W and administrated antioxidant therapy (vit A and glutamine) to reduced or ameliorate such stress. A total of thirty six, male Sprague-Dawley rats weighing (250 – 300) g divided equally into six groups. The first group, control, were fed only standard rat chow as their diet and water ad libitum. The second group administered orally a single dose of methotrexate at a dose of 10 mg/kg B.W. The third group, given a single dose of methotrexate 10 mg/kg B.W + Vit-A (5000 IU/kg B.W) orally by stomach tube, twice daily. The fourth group, given a single dose of methotrexate 10 mg/kg B.W + Glutamine (500 mg/kg B.W), twice daily dosing. fifth group, given vit-A at dose 5000 IU/kg B.W orally, twice daily. Sixth group, given glutamate at dose 500 mg/BW orally, twice daily, all this dosing continue for 8 days. At the end from all animal, draw blood from heart by syring to obtained serum to determine its level then sacrificed and take section from liver for histopathology resulted in. It was found that methotrexate caused a signifcant increase in GSH levels (an important marker of lipid peroxidation) when compared with the control group while the level GSH were signifcantly decreased in the methotrexate + vitamin A group, methotrexate+ glutamine group, vitamin A group and glutamine group. The results indicate that methotrexate cause oxidative stress by reducing the activities and consequently the effectiveness of the antioxidant enzyme defense system. while the antioxidant therapy reducing of oxidative stress. Histopathological examination observed was normal in methotrexate treated rats and MTX with glutamine and vitmine A . but showed hydrodegenerate in treated groups of MTX+vit A while the glutamine treated group showed a necrosis and amyliod change.
