الباحثونRajaa A. Hussein*,.، Nada A. Salih1 and Eman Thabit N
المجلةPlant Archives
مختصر البحث

The present study was carried ot to investigate bioactivity activity of Crocin carotenoid pigment, which obtained from the stigma of Crocus sativus L. plant. It characterized by thin layer chromatography TLC, in UV-visible and IR spectroscopy, antimicrobial activity detected by disc diffusion method and antioxidant activity using DDPH scavenging assay. The results of Crocin extract showed significant inhibitory activity against selected strains of bacteria (Staphylococcus aureus and Escherichia coli) with inhibition zones values ranging from 20 to 36 mm at the concentration of 100 μg/ml as compared with standard antibiotics chloramphenicol and ciprofloxacin. Crocin pigment exhibited appreciable in vitro antioxidant activity (at a concentrations of 50-400 μg/ml )as assessed by 1, 1- diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging as compered with ascorbic acid. It had free radical scavenging capacity at 300 μg/ml (65.7%, IC50 202 μg/ml) close to that of the positive ascorbic acid drug (71%, IC50 178 μg/ml) in the DDPH assay. Key words : Antimicrobial activity, DDPH antioxidant activity assay, crocin, saffron plant.

مختصر البحث

Various assays are used to determine the toxic effects of drugs at cellular levels in vitro. One of these methods is the dye exclusion assay, which measures membrane integrity in the presence of Trypan blue. Trypan blue the dye which was used in this study to investigate cytotoxic effect of a new Cis –dichloroplatinum (II) complex [(Qu)2PtCl2] on the viability of polymorphonuclear cells (PMNs). Three concentrations of platinum complex were prepared (70, 35and 17.5 μg/ ml) and the results revealed that the percentage of cell viability decreased as the platinum complex concentration increased in comparison with control. The platinum complex exhibited low cytotoxic effects towards healthy cells at the concentrations of 17.5 μg/ ml and 35 μg/ ml, in which the percentage of cell viability was (77.01 ± 6.3) and (72.3± 0.50)respectively, with no significant differences as compared with the control(90.66 ±0.577). The viability was significantly decreased (67.59 ± 3.16) when the cells were treated with the concentration of 70 μg/ ml in comparison with control. These results indicated that the percentage of living cells decreased when treated with high concentrations of [(Qu)2PtCl2], which causes cells death, while low concentrations of the compound show low toxicity. This data indicates that this compound, at these concentrations may be suitable for use as a cancer treatment because it has low toxic effects on the healthy cells.

مختصر البحث

Multidrug resistant microorganisms are globally becoming a major confrontment because of illogical use of antibiotics and this played a good role in investigation about the antibacterial compounds in plants. Thus, the present study investigate for the antibacterial effect of alcoholic extracts of Curcuma longa L. rhizomes , CommiphoramyrrhaL. gums and Ginkgo biloba L. leaves products against Staphylococcus aureus and Pseudomonas aeruginosa. The plants samples extracted by soxhlet with methanol and fractionation with and four solution ( chloroform, hexane, water and ethyl acetate) were used for investigation about antibacterial activity by disc diffusion method. The results showed that methanolic alcohol extract and fractions of C. longa L. rhizomes , C. myrrha L. gums showed biological activity against P. aeruginosa and S. aureus bacteria, but methanolic alcohol extract and fractions of G. biloba L. leaves product didn’t show any activity as antibacterial substance. It can be concluded that the presence of secondary metabolites as flavonoids, alkaloids, tannins, glycosides and saponins in the plants under study would be marked a good anti-bacterial effect.

مختصر البحث

Back ground: the plants importance resulting from their uses as a nutrition source and for their therapeutic effect. These plants include turmeric , myrrh and ginkgo. Aim of study: this study was conducted to test the anti-inflammatory effectiveness of turmeric plant ( Curcuma longa L. ) rhizomes and myrrh (Commiphora myrrha L.) gums and ginkgo , Ginkgo biloba L. leaves ( tablets )extracts. Method: plants under study were extracted by soxhlet using methanol as a solvent, and then was fractioned by water, chloroform, ethyl acetate and hexane. The extracts tested to determine the anti-inflammatory effect through testing their ability to maintain the stability of the membranes of red blood cells , prevent protein denaturation and heat induced hemolysis. Results: the result showed the effectiveness of all the plants under study in resisting inflammation with compared to standard medicines (Aspirin 100μg / ml and Diclofenac sodium 100μg / ml). Conclusion: It can be concluded that the presence of secondary metabolites as alkaloid, polyphenolic compounds and phenolic acid in the plants under study would be marked a good anti-inflammatory effect.

Abstract: The use of plant extracts is very important and significant in therapeutic treatments that are employed to many functions in view of the fact that primeval epochs. The present study was conducted to investigate antioxidant effect of extracts from some medicinal plants, extracted by soxhlet with methanol alcohol and fractionation of active constituents from Curcuma longa L. rhizomes , Commiphora myrrha L. gums and Ginkgo biloba L. leaves with water , chloroform , ethyl acetate and hexane. Investigation of the antioxidant activity carried out by DPPH Free Radical Scavenging Assay and showed that all plants were significant (p ≤ 0.05) antioxidant agents compared to ascorbic acid.

The medicinal plants contain many ingredients which enhance the development and synthesis of many drugs the ingredients content of medicinal plants that have therapeutic activity naturally synthesized and accumulated in plants. The presence study was conducted to investigate in vitro and in vivo cytotoxicity effect of some medicinal plants, extracted by soxhlet with methanol alcohol and fractionation of active constituents from Curcuma longa L. rhizomes , Commiphora myrrha L. gums and Ginkgo biloba L. leaves with water , chloroform , ethyl acetate and hexane. The methanolic alcohol extract and fractions of Curcuma longa L. rhizomes , Commiphora myrrha L. gums and Ginkgo biloba L. leaves show ( in vitro ) relatively significant (p ≤ 0.05) low cytotoxicity against RBC , while ( in vivo ) the cytotoxicity at 2000 mg / ml did not occur which indicated the study plants safety.
