Abstract

ABSTRACT The principal objectives of this research were to prepare and evaluate Meloxicam nanoparticles loaded fast dissolving film, and nanoparticles were used to permit for enhancing dissolution of the drug, while the films were used to fasten the onset of action Meloxicam nanoparticles were fabricated by solvent/antisolvent method and then formulated as the oral thin films by casting method. The Meloxicam nanoparticles were prepared at selective polymer: drug ratios of 1:1, 2:1, and 3:1 using various polymers and specific grades of polyvinyl pyrrolidone and hydroxy-propyl methyl cellulose as stabilizers. The effect of polymer, ratio, and stirring rate on the size of the particulate, were studied, and found to have a significant (p ≤ 0.05) effect. The best formula (FMx1) was obtained with a minimum average particle diameter of 59.5 nm and surface area 37.34 m2/g contained 1:1 of HPMCE-50LV: Meloxicam. This formula was freeze-dried and studied for surface morphology by Field Emission Scanning Electron Microscope (FESEM), crystalline state by XRPD, and compatibility by FTIR and DSC. Then Meloxicam nanoparticles were formulated into an oral thin film. The obtained film Meloxicam content of 7.4 ± 0.02 mg per 4 cm2 of film and disintegrated in 29 seconds. The Meloxicam nanoparticles film was illustrated a faster in vitro dissolution rate than the film prepared from pure Meloxicam, and the FESEM images indicated that Meloxicam nanoparticles were greatly distributed into the films. These results indicated that the oral film prepared from Meloxicam nanoparticles could be a favorable system to improve the dissolution and fasten the action. Abstract

Background: Ibuprofen is a nonsteroidal anti-inflammatory drug (NSAID) having poor water solubility and high permeability. The study aimed to prepare, characterize, and evaluate ibuprofen nanosuspensions to enhance its solubility and dissolution rate. Materials & Methods: This was an experimental and laboratory-based study. Nanosuspensions of ibuprofen were formulated using three different polymers-PVP K30, Poloxamer 188, and HPMC E50-at various drug-to-polymer ratios (1:1, 1:2, 1:3). The study was conducted in a controlled laboratory environment, typically in a pharmaceutical or chemical research laboratory equipped with instruments like particle size analyzers, scanning electron microscopes, and dissolution apparatus. The formulation and characterization experiments were performed in compliance with standard protocols for nanoparticle formulation and pharmaceutical analysis Results: All ibuprofen nanosuspensions were within the nano size range (123-564.5 nm), with the smallest particle size (123 nm) observed for PVP K30 at a 1:1 ratio. The PDI ranged from 0.008 to 0.071, and the specific surface area (SSA) ranged from 18.6 to 3.39 m²/g. Saturated solubility of ibuprofen increased by approximately six-fold compared to the pure drug. Entrapment efficiency ranged from 85% to 97%, while the dissolution rate showed a significant improvement, with 98% of the drug released within 15 minutes, compared to 25% release from pure ibuprofen in the same period. Conclusion: The solvent- Anti-solvent technique is effective for producing ibuprofen Nano-suspensions, significantly enhancing the solubility and dissolution rate of the drug. This formulation approach holds promise for improving the bioavailability of poorly water-soluble drugs like ibuprofen.

Abstract

Nanoparticle technology is currently widely employed to accelerate drug dissolving and greatly improve bioavailability. Nanoparticles were mixed with hydrogel to create a nanoparticle-hydrogel hybrid system as an alternative to traditional dosage forms. The purpose of this work is to create an effective hydrogel form containing ketoprofen nanoparticles for topical application, ketoprofen a potent non-steroidal anti-inflammatory drug. Eleven nanoparticle formulations of ketoprofen were prepared and characterized for particle size, specific surface area SSA and PDI, by use different polymers (PVP-K15,HPMC-E15 and HPMC-E50) in three different polymer: drug ratio (1:1,2:1 and 3:1).The results indicate increasing in the concentration of PVP K15 generally reduces significantly (p≤0.05) ketoprofen particle size, so select the smallest size formula F3 contain PVP-K15 in ratio polymer :drug (3:1) was 74.9 nm for further characterization which are SEM, X-ray and DSC ,then use this F3 formula to prepare hydrogel and assessment the physical appearance, pH, drug content, spreadability test, viscosity, and in-vitro drug release. The results showed the drug content of ketoprofen nanoparticles F3 in hydrogel was 96.4%, spreadipility was 11.2 gm.cm2/sec, hydrogel viscosity was (6500cps to 75400cps), and drug release from F3 hydrogel was 96.13% at 10hr. there was a significant enhancement in drug release from this nanoparticle hydrogel than plain ketoprofen hydrogel.

Abstract

Topical drug administration is a technique of applying different dosage forms, including ointments, creams, hydrogels, to the skin and different sections of the body, it has systemic or local effect. Nimesulide is a non-steroidal anti-inflammatory medication with strong analgesic, and anti-inflammatory properties, it was used in this research and prepared as nanoparticles, which was then incorporated into hydrogel for topical application, to increase solubility of Nimesulide, which is class II in BSC, and reduce their systemic side effects. Nimesulide nanosuspension were prepared by solvent/antisovent method at different polymer: drug ratios (1:1, 2:1, and 3:1) using polymers as polyvinyl pyrrolidone (PVP-K15, PVP-K30), hydroxypropyl methylcellulose (HPMC-E5) and poloxamer-188. The effect of polymer type and concentration on the particle size, poly dispersity index, specific surface area was studied. Among twelve prepared formulas, F2 was selected as best formulas with an average particle size of 59. 5 nm. It was lyophilized and investigated for surface morphology by FESEM, drug–excipients compatibility studies by DSC, crystalline state by XRPD and then Nimesulide nanosuspension of F2(1%w/w Carbopol) was formulated into hydrogel dosage forms and subjected for further evaluation. The results indicated that particle size of nanosuspension was found to be affected by type and concentration of polymer and when increase polymer: drug more than 1:1 led to increase particle size. In addition, the release profile of F2 hydrogel formula was better than plain Nimesulide hydrogel. From this, it is concluded that the dissolution rate of Nimesulide nanosuspension was increased through particle size reduction to nanometer size.
