The aim of the study is to measure the concentration IgG antibody of the human vaccinated, which include (Pfizer, AstraZeneca, and Sinopharm vaccinations) and to find out which vaccine is more efficient by RT-qPCR technology. It highlights risk factors like age and sex. The cross-sectional study took place between November 25, 2021, and January 25, 2022. Background: Studying the immune response to different COVID-19 vaccines is crucial for refining vaccine selection and dosage worldwide. The aim of the study is to measure the concentration IgG antibody of the human vaccinated, which include (Pfizer, AstraZeneca, and Sinopharm vaccinations) and to find out which vaccine is more efficient by RT-qPCR technology. It highlights risk factors like age and sex. The cross-sectional study took place between November 25, 2021, and January 25, 2022. Results: In our study of eighty-six cases, fifty tested positive for COVID-19, comprising thirty-eight females and twelve males. Monitoring occurred from the second to the seventh-month post-vaccination, with IgG titers measured via RT-qPCR. We observed higher vaccine efficacy in females compared to males. Age groups were categorized as follows: 19-29, 30-40, 41-51, and 52-62 years old. The 19-29 age group showed the highest proportion vaccinated with Pfizer, AstraZeneca, and Sinopharm vaccines. IgG concentration after 7 months was significantly higher than in earlier months. Further, IgG levels were lower in participants vaccinated with Sinopharm (p<0.0001) and AstraZeneca (p<0.01) compared to Pfizer. Conclusion: Participants who received the Pfizer vaccine demonstrated the highest antibody concentration relative to other vaccine recipients. There was observed higher vaccine efficacy among females. Notably, the 19-29 age group represented the largest proportion of individuals vaccinated with various vaccines. Consequently, this study holds significant potential for optimizing vaccine selection and dosage. Further investigations are warranted to delve deeper into these initial findings.

Gluten-appropriate immune response is a key feature of an a… Abstract Background: The current research examines the molecular terrain of celiac disease (CD) through microRNA (miRNA) and cytokines as potential new diagnostic and therapeutic markers. Gluten-appropriate immune response is a key feature of an autoimmune clinical entity known as CD that leads to inflammation and degeneration of small intestine mucosa. However, the mechanisms responsible for this remain unclear. Methods: Quantitative reverse transcription polymerase chain reaction (RT-qPCR ) was carried out on serum samples obtained from patients with CD and control groups to unravel their pathogenesis. Assessing miR-155, miR-15b, interleukin (IL)-2, IL-7, IL-35and IL-37 levels in expression might be useful in diagnosing or treating the disorder. Results: A significant dysregulation of these molecular players in patients with CD compared with healthy controls has been evidenced by results from this study. For instance, miR-155 was up-regulated, whereas miR-15b was significantly down-regulated in CD, illustrating their roles in immune responses and inflammation-mediated processes. Besides, there was an over-expression of IL-2 and an under-expression of IL-37 in patients with CD, indicating these biomolecules’ role in immuno-dysregulation and inflammatory process underlying CD. In addition, a positive correlation between IL-2 and miRNA 155 expression levels was observed in patients with CD, suggesting that they could be involved together with other cytokines, showing the interplay between immune response pathways and inflammatory cascades during CD pathogenesis. Conclusion: These molecular signature discoveries might result in new and revolutionary diagnostic modalities and moleculartargeted therapies for CD pathogenesis. When used with the scientific understanding of miRNAs and cytokines associated with CD pathophysiology, it creates a basis for personalized medicine based on the individualized molecular profile of all patients. This will undoubtedly increase the efficacy of CD treatment strategies. In brief, more research on molecular pathways’ workings should be done to harness their potential in CD diagnosis and treatment.

These studies have primarily now concentrated on the microbial communities that… Background In human and animal studies, ankylosing spondylitis (AS) has been increasingly linked to changes in the microbial inhabitants in the human body (microbiome). These studies have primarily now concentrated on the microbial communities that live in the gastrointestinal tract. However, evidence suggests that various molecular techniques can be used to detect microbial DNA in blood circulation. This DNA might be an unknown reservoir of biomarkers with the potential to track alterations in the microbiomes of remote locations, such as the gut. To this end, we compared the presence and identity of microbial DNA in blood samples taken from ankylosing spondylitis patients to healthy control subjects by amplifying and sequencing the bacterial 16S rRNA variable region four. Methods The study's design is a case study based on the presence and identity of bacterial DNA in the blood of Ankylosing spondylitis (AS) patients (n = 10) and healthy control subjects (n = 10) was investigated by amplifying and sequencing the bacterial 16S rRNA gene. Blood concentrations of the cytokines TNF alpha, IL-17A, and IL-23 were determined by the Human Magnetic Luminex Screening, and data were analysed using an Unpaired T-test. Results Using PCR amplification, 8 of 10 AS patients (80%) and 8 of 10 healthy control samples (80%) had microbial 16S rRNA in their blood. At the phylum level, Proteobacteria (Control = 48.5%, AS = 52%), Firmicutes (Control = 27.8%, AS = 26.1%), Actinobacteria (Control = 15.4%, AS = 10.7%), and Bacteroidetes (Control = 6.5%, AS = 10%) dominated the blood microbiome. A two-tailed Mann-Whitney test found that Ankylosing Spondylitis was associated with significantly elevated Bacteroides (P < 0.05), Prevotella (P < 0.001), and Micrococcus (P < 0.01), and significantly reduced levels of Corynebacterium 1 (P < 0.001), Gemella (P < 0.01), and Alloprevotella (P < 0.05), compared to healthy controls. Additionally, it was shown that the presence of the Prevotella genus was highly positively correlated with higher levels of TNF-alpha (P < 0.05; r = 0.8) in AS patients' blood. Conclusion This article reveals that a blood microbiome exists in healthy individuals and identifies particular taxa modulated in disease. These blood-derived signatures indicate that this field needs more research and may be helpful as disease biomarkers.
