This study aimed to investigate the distribution of uropathogenic Escherichia coli
(UPEC) associated with urinary tract infect. A total of 150 urine sample have been
collected from patient suffering from UTI whom add minted to Al_Najaf Al_ashraf
hospitals from both gender (female and male ) involved 107 urine samples from female
with age group range between 14-50 years and 43 urine samples from male with age group
range between 8-45 years . The results of microscopic, phenotypic and biochemical test
showed that only 30 isolates were belong to UPEC in which 24 (22.42%) isolates from
female and 6(13.95%) isolates from male.
PCR technique has been used for detection the serotype of UPEC by using O15 and
O25 serotype gene and the results of gel electrophoresis showed that only 5(16.66%)
isolates were belong to O15 by appearing of amplicon with 183 bp while 6(20%) isolates
were belong to O25 with 230 bp of amplicon .
The result of agarose gel electrophoresis for detection presence of some virulence
gene showed that the gene product of cnf1, pap, fim, sfa, papGII, iha and iss showed that
only 3(10%) isolates have cnf1 gene with 498 bp amplicon while 8(26.6%) isolates,
8(26.6%) isolates, 12(40%) isolates, 7(23.3%) isolates, 18(60%) isolates and 19(63.3%)
isolates were possess fim (559 bp), pap (336 bp), papGII (190 bp), sfa (410 bp), iha (827
bp) and iss ( 309 bp) respectively. Also the results revealed that only 2 (6.66%) isolates of
UPEC were possess all these genes while 6 (20%) isolates have only 3 different genes
where 7(23.33%) isolates did not possess any one of studied gene.
