Introduction: Luteolin is highly distributed within plant species it is approved to be efficacious in the treatment of ischemic stroke caused by inflammation. Ischemic stroke (I/R) damage to the brain is a commonly diagnosed natural outcome of ischemic stroke. The objectives of this paper are to use in silico molecular modelling to evaluate the primary biomarker proteins with which luteolin binds, as well as the benefits of luteolin in minimizing the harm caused by cerebellar (I/R) injury. Methodology: The molecular modelling research was performed using AutodockFr. The Bcl-2, Caspase-3, IL-1B, MDA, TLR-2, and TNF- values were observed, alongside the histological assessment which was used to determine the level of restoration from the cerebellar (I/R) injuries sustained in mouse models. Results: Docking studies demonstrated that the luteolin structure can interact with both SOCS3/JAK and SYK Enzymes. Concomitantly, the physiological responses showed marked declines within Bcl-2, Caspase-3, IL-1B, MDA, TLR-2, and TNF- brain chemicals, with huge rises observed with Bcl-2 concentrations. Moreover, the histopathological examination revealed that luteolin decreases ischemic injury. Conclusions: Lutein could be able to decrease cerebral (I/R) damage by decreasing oxidative stress and trying to exert an anti-apoptotic overall impact.

مختصر البحث

The food industry has long recognized nanoparticle delivery for uses ranging from slow-release vitamin and mineral formulations to nutritional supplement. Small particles that can be easily dissolved are used to create a bioavailable formula or add vitamins and minerals to a “clear” food beverage thus helping fat-based food products taste better. The colloidal delivery strategy (primarily dealing with micro and nanoencapsulations) has been deemed a priceless resource for resolving many issues involved in food fortification. Encapsulated minerals can be applied and integrated into the formulation of food and medicine to provide unique benefits for the development of physically and chemically improved functional products. In this chapter, we explain different nano and microencapsulations of vitamins and minerals and how food fortification can be used as a low-cost solution to vitamins and minerals deficiency.

مختصر البحث

The research aimed to investigate the anti-inflammatory effect of 4-hydroxy coumarin derivatives on inflammatory response in rats and to estimate the toxicity of enumerated compounds (3-11) following acute exposure. Toxicity report was collected by oral dosing administered individually to rats on a scale of 10-3000 mg/kg, while an indication of toxicity was observed for ten days following the last given dose. The response to inflammation was estimated using the model of xylene-induced edema and the model of carrageenan-inflammation of the paw. Compounds were administered orally to animals at a once-daily dose for seven sequential days. Molecular docking studies with COX-II enzyme were used to explain deeply the activity of synthesized compounds. Furthermore, the in-silico results were quite consistent with biological activity. Interestingly, compounds 5 and 10 showing the highest binding points (lower values). Therefore, the molecular dynamics simulation study of the most active compounds 5 and 10 was performed.

SARS-CoV-2 is a new generation of coronavirus, which was first determined in Wuhan, China, in December 2019. So far, however, there no effective treatment has been found to stop this new generation of coronavirus but discovering of the crystal structure of SARS-CoV-2 main protease (SARS-CoV-2 Mpro) may facilitate searching for new therapies for SARS-COV-2. The aim was to assess the effectiveness of available FDA approved drugs which can construct a covalent bond with Cys145 inside binding site SARS-CoV-2 main protease by using covalent docking screening. We conducted the covdock module MMGBSA module in the Schrodinger suite 2020-1, to examine the covalent bonding utilizing. Besides, we submitted the top three drugs to molecular dynamics simulations via Gromacs 2018.1. The covalent docking showed that saquinavir, ritonavir, remdesivir, delavirdine, cefuroxime axetil, oseltamivir and prevacid have the highest binding energies MMGBSA of –72.17, −72.02, −65.19, −57.65, −54.25, −51.8, and −51.14 kcal/mol, respectively. The 50 ns molecular dynamics simulation was conducted for saquinavir, ritonavir and remdesivir to evaluate the stability of these drugs inside the binding pocket of SARS-CoV-2 main protease. The current study provides a powerful in silico results, means for rapid screening of drugs as anti-protease medications and recommend that the above-mentioned drugs can be used in the treatment of SARS-CoV-2 in combined or sole therapy.

The present work pointed at the design and synthesis of novel coumarin compounds as antioxidants. The alteration of 4-hydroxycoumarin by several reaction levels was performed to generate destination compounds. Spectroscopic methods like Infrared Spectra FT-IR, 1H-NMR and 13CNMR, elemental analysis techniques, melting point, and thin layer chromatography defined the structure of the prepared compounds. The antioxidant activities of individual compounds were tested against stable free radical 1,1-Diphenyl-2-picrylhydrazyl radical (DPPH), hydrogen peroxide (H2O2) and FRAP (ferric reducing antioxidant power) and the results were related to the Gallic acid, ascorbic acid and Trolox as standards. The effects revealed that most of the syntheses presented greater activity as an antioxidant than the standards in different concentration range. The distinguished competence inactivating action was observed for compound 2 (86.3±1%) followed by compounds 3 (85±0.5%) using DPHH method. The antioxidant mode of action of the prepared compounds was also investigated.

ABSTRACT : Isatin was first synthesized in 1841 by Erdmann and Laurent by the oxition reaction of indigo dye by an oxiizing agent nitric acid/chromic acid. It was found in many plants, like Isatis tinctoria, Calanthe discolor and in cannonball tree. Isatin- azomethine had been studied for their important medicinal applications. The syntheses of Isatin- azomethines, that are quite compulsory pharmaceutical properties because of their structurally motivating antioxidant activities. Isatin - azomethines had been synthesized effectively via the reaction of Isatinwithvarious amins. Various spectroscopicaltechniqes were used to characterized the synthesized Isatins. The antioxidant capacity of Isatin-azomethines were predestined by representative spectroscopically techniques, employing radical’s DPPH (2,2-diphenyl-1- picrylhydrazyl), FRAP (Ferric Reducing Antioxidant Power) and Ferrous ions chelating activity assays compared to that of ascorbic acid. Isatin-azomethines displayed an superior scavenging activities paralleled to the ascorbic acid.

Background: Coumarin is quite extend in plants including various vegeta- bles, flavors, foods. Coumarin has various significant medicinal activities such as anti-inflammatory, anti-oxidant, anti-viral, anti-microbial, anti-cancer and has been indicated to increase central nervous system activity. Methods: The syntheses of tailor-made coumarins, which are highly compelling for medical applications due to their structurally interesting antioxidant activity, were report in this manuscript. Coumarins were synthesized success- fully through the modification of 4-hydroxycoumarin by different reaction steps. The molecular structures of the coumarins were characterized by the Fourier transformation infrared and Nuclear magnetic resonance (NMR) spectroscopies. The antioxidant efficiency of the 4-hydroxycoumarins were evaluated by typical spectroscopic method, using radicals DPPH• and H2O2. Results: The new coumarins synthesized in this work exhibited an excellent antioxidant compared to the free vitamin C. Molecular modelling studies using DFT (Density Functional Theory) calculations showed that there is a high correlation between dipole moment, Ionization potential (IP), Electron affinity (EA), Hardness (η), Softness (S), Electro negativity (μ), energy gap, HOMO (Highest Occupied Molecular Orbital) and LUMO (Lowest Unoccupied Molecular) energies and antioxidant activity. Conclusion: New coumarin derivatives were successfully synthesized using chemical methods. The characterized of these coumarins were done by using different spectroscopic techniques (FT-IR and NMR) and micro-elemental analysis (CHNS). The antioxidant activity of these coumarins were determined by using DPPH and hydrogen peroxide assays. Results indicated that the new coumarins possess higher scavenging activity than vitamin C.

Compounds that can bind and stabilize non-canonical DNA structures are named quadruplex and are of interest in anticancer drug design due to their selective inhibitions of telomerase and consequent effects on cell proliferation. In this study, we report novel Co/Cu [II] complex compounds as G-quadruplex DNA binding ligands. The results from the preliminary assay indicated that the introduction of a positively charged 6-membered tail to the aromatic terminal group of benzimidazole significantly enhanced the binding affinity with the quadruplex and exhibited anti-telomerase activity. These derivatives showed significant selectivities for the telomeric quadruplex over duplex nucleic acids. The stabilization of non-canonical forms estimated with the FRET DNA technology using different sequences, such as F21T, c-kit1 and c-kit2, in cancer cell lines were assessed. Three members of this family showed to be very selective in stabilizing one particular G-quadruplex.

Syntheses of coumarins, which are a structurally interesting antioxidant activity, was done in this article. The modification of 7-hydroxycoumarin by different reaction steps was done to yield target compounds. Molecular structures were characterized by different spectroscopical techniques (Fourier transformation infrared and nuclear magnetic resonance). Antioxidant activities were performed by using various in vitro spectrophometric assays against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydrogen peroxide (H2O2). All compounds exhibited high efficiency as antioxidants compared to ascorbic acid. The highest efficiency scavenging activity was found for compound 3 (91.0 ± 5.0), followed by compounds 2 and 4 (88.0 ± 2.00; and 87.0 ± 3.00). Ascorbic acid C was used as a standard drug with a percentage inhibition of 91.00 ± 1.5. The mechanism of the synthesized compounds as antioxidants was also studied. Hartree–Fock–based quantum chemical studies have been carried out with the basis set to 3-21G, in order to obtain information about the three-dimensional (3D) geometries, electronic structure, molecular modeling, and electronic levels, namely HOMO (highest occupied molecular orbital) and LUMO (lowest unoccupied molecular orbital), to understand the antioxidant activity for the synthesized compounds.

Introduction: Coumarin is a natural product compound known for its medicinal properties. Methods: The rational design of 4-hydroxycoumarin functionalized thiadiazoles (3 and 4) was synthesized with tailor-made antioxidant properties. The strategy involves intra-molecular cyclization of 2-(coumarin-4-yloxy) acetic acid under reflux conditions. Antioxidant activity of synthesized compounds were performed using various in vitro assays against 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and hydrogen peroxide (H2O2) scavenging. Results: The results revealed that tested compounds 3 and 4 are much higher than well-known antioxidant compound naming ascorbic acid. They showed good scavenging capacity against DPPH and hydroxyl radicals. Ab initio HF (Hartree–Fock) with 3-21G basis set for the title compounds demonstrated the correlation of scavenging activities and theoretical parameters (such as, dipole-moment, ionization-potential, electron-affinity and highest occupied molecular orbital). Conclusion: In conclusion Coumarins were successfully synthesized and were evaluated as antioxidants by DPPH and hydrogen peroxide assays and they were indicated that they had good scavenging activities. The synthesized antioxidants were investigated theoretically and also some quantum chemical parameters were calculated.

مختصر البحث

Recently non-steroidal anti-inflammatory drugs (NSAIDs) have been proposed to prevent or to cure Alzheimer’s disease. In this respect, we synthesized new potential prodrugs of several NSAIDs in order to increase their access to the brain. The carboxylic group of NSAIDs was attached to the 1,4- dihydro-1-methylpyridine-3-carboxylate moiety, which acts as a carrier, via an amino acid bridge, according to the chemical delivery approach developed by Bodor. In this study new analogues were synthesized by insertion D- Threonine moiety using esterification method then linked to nicotinic acid by conventional solution method using DCC as coupling reagent all prodrugs were more lipophilic when compared to their corresponding parent compounds and consequently a better blood brain barrier (BBB) penetration is hypothesised the analogues were purified and monitered by TLC , melting point , and IR spectrophotometer FT.IR 8400 was achieved and CHNS analysis.
