Abstract

This study presents a new, straightforward, and precise spectrophotometric technique for determining the amount of paracetamol. The azo dye is formed using a new chromogenic reagent, orcinol, to produce an orange-reddish result that is detected at 514 nm. UV-visible and other spectroscopy methods were used to confirm the newly synthesized compound's molecular structure. The azo dye derivative produced was tested against four bacterial species in vitro. A linear connection between the concentration range of 0.8-20.5 μg. mL-1 of paracetamol was achieved under carefully controlled experimental conditions that impact the response. In addition to the molar absorptivity of 12.54×106 L.mol-1.cm-1, the regression coefficient (R2) was 0.9961. For the purpose of determining paracetamol in the pharmaceutical business, the suggested methodology was successfully adjusted

Indirect Spectrophotometric Technique for Quantitative Evaluation of Some Phenolic Medicines

AuthorsBasim F. Moammed AliI
JournalUniversity of Kufa
Abstract

This study introduces a novel, cost-effective, and rapid indirect spectrophotometric method for the quantification of three phenolic pharmaceuticals: Doxycycline hyclate (DOX), Cefadroxil monohydrate (CFL), and Oxymetazoline hydrochloride (OMN), in both pure and dosage forms.1. Methodology and Reaction MechanismThe proposed analytical technique is based on a redox-bleaching reaction involving three primary steps:Oxidation: The drug is oxidized in an acidic medium using a known excess of N-Bromosuccinimide (NBS).Dye Interaction: The remaining (unreacted) NBS reacts with Methylene Blue (MB) dye.Quantification: The concentration of the drug is determined indirectly by measuring the absorbance of the residual MB dye using UV-Visible spectrophotometry at a maximum wavelength ($\lambda_{max}$) of 580.0 nm.2. Optimization of Experimental ConditionsThe study established specific optimal parameters for each drug to ensure maximum sensitivity:Reagents: Nitric, acetic, and hydrochloric acids were identified as the most effective media, respectively.Order of Addition: For DOX and OMN, the sequence follows: Acid → Drug → Oxidant → Dye. For CFL, the optimal sequence is: Drug → Acid → Oxidant → Dye.Thermodynamics & Kinetics: Optimal temperatures ranged between 5 °C and 20 °C. Oxidation and bleaching times were meticulously timed to ensure complete reaction before measurement.3. Analytical Performance and ValidationThe method demonstrated high statistical reliability and adherence to international validation standards:Linearity: All drugs followed the Beer-Lambert law within concentration ranges of 5–25 µg/mL, with high correlation coefficients ($R^2 > 0.998$).Sensitivity: The method achieved low Detection Limits (LOD) and Sandell’s sensitivity values, indicating high precision for trace analysis.Accuracy and Precision: Relative Standard Deviation (RSD%) and Recovery (Re%) values (averaging ~100%) confirm the method's reproducibility and exactness.4. Pharmaceutical Application and Comparative AnalysisSelectivity: The method was successfully applied to various commercial formulations (capsules and nasal sprays) available in Iraqi pharmacies. Common excipients showed negligible interference (less than 2%).Statistical Significance: Comparisons between the proposed method and standard reference methods using t-tests and F-tests showed no significant differences, proving that this low-cost method is a viable alternative to highly specialized equipment. Conclusion: The developed method provides a highly selective, precise, and accessible alternative for the quality control of phenolic medicines without the need for sophisticated instrumentation.

Utilization of Spectrophotometric Technique for the Quantitative Evaluation of Doxycycline in its Pure form and its Therapeutic Doses

Abstract

A rapid and easy spectrophotometric system has been proposed to determine Doxycycline Hyclate (DOX) in pharmaceutical formulations. The oxidation strategy consists of putting Doxycycline with an excess quantity of N-Bromo succinimide (NBS). That happens in an acidic medium and then estimates the amount of unreacted NBS by methylene blue at λ580 nm. The quantity of NBS interacting with the colourant is equivalent to the amount of the drug with a better molar absorption of 2.44 × 103 L. mol-1 cm-1. The bear law scale was 5-25 µg.ml-1, and Sandell's sensitivity index of 0.210 g.cm-2. And also, the average relative error of the concentration is -0.503 %, and the relative standard deviation is 0.337%. The method was successfully applied in its quantifying to the determination of drug in pharmaceutical formulations.

Abstract

Establishing and adopting a quick, easy, and cheap method for determining antibiotic Cefadroxil monohydrate (CFL) in its pure form and pharmaceutical preparations. The method involves adopting spectrophotometry in the visible region. The method adopted the oxidation of the studied medicine by an excess amount of N-Bromo succinamide (NBS), then estimated the NBS Excess by bleaching the dye color of Methylene Blue. The maximum wavelength was measured at λ 586 nm. This approach obeys the Beer-Lambert law for the range (5-20 µg. ml-1), with a correlation coefficient of 0.9980. The calculated molar absorptivity was 1.41× 103 (L. mol-1. cm-1), and Sandall's sensitivity was 0.27 µg. cm-2. The limit of detection (LOD) was 3.508 µg. ml-1, as well as the limit of quantification (LOQ), was 11.693 µg. ml-1. The precision and accuracy of the method are established and checked. This study measured the optimal reaction conditions and other analytical variables. It showed decent repeatability, and the relative standard deviation (RSD percentage) was less than 1%. The proposed methodology identified the effective implementation of an easy, resilient as well as precise spectrophotometric method to determine Cefadroxil employing N-Bromo succinimide as Cefadroxil oxidant factor and also the unreacted N-Bromo succinimide bleached the methylene blue dye. The process has been successfully extended in different pharmaceutical formulations to evaluate the Cefadroxil drug.
