Detection p53 gene deletion in hematological malignancies using fluorescence in situ hybridization: a pilot study

مختصر البحث

Abstract P53 as a tumor suppressor gene plays a major role in cancer development, it is essential for cell growth regulation and apoptosis. The deletion of p53 is known to be associated with aggressive diseases in several hematological malignancies. The evidence indicated that p53 deletions can be acquired as a result of chemotherapy. Therefore, a follow-up study for p53 gene deletion by fluorescence in situ hybridization technique (FISH) was carried out for the patients group who affected with different hematological malignancies before and after chemotherapy. The main goals from screening of p53 deletion were to assess the correlation between p53 deletion and chemotherapy resistance, overall median survival and chromosomal abnormalities. It is concluded from the present study that p53 deletion has a cardinal effect on the clinical outcome (chemotherapy resistance, overall median survival) and outcome of chromosomal abnormalities (quality and quantity of chromosomal abnormalities) of the patients who were affected with hematological malignancies before and after chemotherapy.

Human Chromosome Variation: Heteromorphism and Polymorphism

الباحثونHerman E. Wyandt, ، Vijay S. Tonk
المجلةSpringer Science & Business Media
مختصر البحث

Human Chromosome Variation: Heteromorphism and Polymorphism was formerly printed under the title “Atlas of Human Chromosome Heteromorphism”. The Atlas has become a standard reference book in most cytogenetic laboratories and is cited as a significant reference in ISCN 2009. This revised version has updated and retained the most useful pictorial sections of the first edition, including the comprehensive review of normal and “not-so-normal” variations of the human karyotype with summaries and extensive reference lists organized by chromosome number. This updated edition features concise background information on chromosome methods and applications, essential information on heteromorphism frequencies in normal and clinical populations as well as new listing and discussions of euchromatic, subtelomeric and FISH variants. The addition of two new sections make this an even more valuable reference than before. A section on common and rare fragile sites includes a short historical discussion, definitions and an extensive table of officially recognized sites that includes the HUGO name, chromosomal location, methods of induction, genes and references to the most recent molecular characterization. A new section on array CGH discusses the clinical challenge of interpreting copy number variations (CNVs) revealed by this newest technology, gives examples of various levels of interpretation and lists the several most common websites used in this interpretation.

Genetic Disorders Among Arab Populations

مختصر البحث

Arab populations have their “own” genetic disorders, both universal and particular. Genetic diversity within these source populations, along with the fact that the rates of inbreeding are often high and family sizes are often large, constitute conditions that facilitate the emergence and detection of phenotypes explained notably by autosomal recessive inheritance; in which case, the use of homozygosity gene mapping can facilitate the discovery of the corresponding genes. The present book includes 5 parts dealing with various aspects that relate to the

High frequency of satellite association in metaphases of infertile male patients

مختصر البحث

Abstract Objective: The present study is designed to evaluate the contribution of satellite association phenomenon on spermatogenic impairment. Methods: During a one year period, January 1999 through to January 2000, the frequency of satellite association has been investigated in 57 patients, with the clinical diagnosis of male infertility. This study was carried out at the Middle Euphrates Center for Cancer Researches, College of Medicine, Kufa University, Kufa, Iraq. Of those 57 patients, 33 patients were diagnosed with azoospermia and 24 patients with oligospermia, the efficacy evaluation was based on the measurement of 8 satellite association parameters. Blood culture and chromosome harvesting was carried out according to our standard methods. Results: Our experience of association behavior of acrocentric chromosome in the 57 infertile males showed statistically significant difference in infertile male classes compared to control groups. Eight studied parameters were included in the evaluation (P<0.005). Conclusion: The significant increase in the satellite association is proposed to have another indirect causal factor, which influenced spermatogenesis. Furthermore, the satellite association technique may be used as a laboratory method for the evaluation of male infertility.

مختصر البحث

Abstract OBJECTIVE: The goal was to estimate the contribution of chromosome anomalies in the Iraqi infertile males. METHODS: Sixty-four male patients were included in the present study. Blood culture and chromosomal harvesting were conducted according to standard methods. RESULTS: The percentage of normal karyotype was 87.5%. The number of abnormal karyotypes constitute about 12.5%. In our azoospermic patients, about 11% of patients stated to have abnormal karyotype comparing to 15% in oligospermic patients. On the other hand, sex chromosomal anomalies were detected in 4 patients with azoospermia. No autosomal anomalies were found in this group. Meanwhile, 3 patients with sex chromosomal anomalies were recorded in oligospermic patients. The unique autosomal anomaly was detected in one oligospermic patient. CONCLUSION: Karyotyping of subfertile males will still be important not only from a diagnostic viewpoint, but even more importantly, in order to gain a better understanding of gametogenic impairment, which is associated with chromosomal abnormalities. Moreover, the value of cytogenetic screening is emphasized since this group of chromosomally abnormal patients can be excluded from conventional treatment.
