Abstract

Cancer is a major health problem worldwide and is one of the leading causes of death. Preventing tumor development is a significant challenge due to the similarity in cellular mechanisms between cancerous and normal cells; however, biological substances have high efficiency against cancerous cells and low toxicity against normal cells associated with a good prognosis. Therefore, the current study is designed to study the role of Anise extract (A.E.) and Anise extract- Chitosan nanoparticles in the synergistic effect with Cisplatin in the treatment of tumor-induced in female mice. Materials and methods: Five groups of female mice, six mice for each group, were included to achieve the goal of the present study. These study groups were control, tumor-induced, Cisplatin treated, Cisplatin – Anise extract treated, and Cisplatin- Anise extract- Chitosan nanoparticles treated groups. The tumor size and some biomarkers (IL-33, LRG-1, and CRP) were considered indicators for tumor development and response to the treatment protocol. Results: The present study found that the tumor size in the tumor-induced group reached 33.1 mm after 30 days of inducement, while the tumor size in the treated group was significantly smaller than in the tumor-induced group. The tumor size in the Cisplatin- Anise extract group was smaller than the Cisplatin treated group. Also, the effect of Cisplatin against a tumor increased when combining anise extract with Chitosan nanoparticles. The biomarkers showed that IL-33, LRG-1, and CRP levels were higher in the tumor group than in the control and other study groups. Conclusion: The current study concluded that the anise extract has high effectiveness in reinforcing Cisplatin action against tumor development and, simultaneously, increased their effect by nanoparticles.

Abstract

Diclofenac is a remedy nonsteroidal anti-inflammatory drug (NSAID); that is used for treating the manifestations of rheumatoid arthritis and is being deliberate for forbidding of some types of skin cancer. It plugs releasing substances that cause inflammation and pain. It may also prevent the outgrowth of new blood vessels that tumors need to get bigger. Diclofenac is most potent, so a little amount of diclofenac is needed to relieve pain. Diclofenac is commonly used for arthritis-related joint pain. Twenty white rats were used, the group was divided into four subgroups, each containing five rats for thirty days of oral directing 50, 100 and150 mg/kg daily, with the control group receiving normal physiological saline, the second group given with 50 mg/kg daily of the cure, the third group given with 100 mg/kg daily of the cure, the fourth group was given 150 mg/kg daily of the cure. The rats were slaughtered after the groups received the treatment for one month. in addition to semithin sections for light microscopic examination, followed by statistical studies. This survy, the xeno-biotic is run on the lab-animals top of a period of 15 to 28 days, and the target mainly to pick out short-term toxic effects or injury that might manifest from refined exposure. Duration: Usually scop from 2 to 5 weeks, during the time blood or tissue specimens are taken up to an eye on any changes in physiology or chemical indicator. This allows for the detection of toxic consequence that may come into view only follow up repeated exposure over a short period. (Bemidinezhad, et al. 2023). The study showed that the rat bone marrow was affected adversely via the managing with Diclofenac at 50, 100 and150 mg/kg daily for 30 days in histologic structure and easy to perceive differences were observed after light microscopy was used to examine different sections of bone from control and treated animals. The Diclofenac-treated groups’ bones showed more active haemopoietic tissue (hypercellularity) relative to adipocytes (white spaces). Diclofenac can cause significant but long-term harm. so this attempt was to determine the effect of Diclofenac on many physiological criteria and several histological indicators in the bone marrow of male rats Novelty Statement | This study reveals that long-term diclofenac sodium manifest may cause bone marrow hypocellularity, reducing hematopoietic cells and potentially leading to pancyto-penia, anemia, and increased infection risk—a drug-induced effect not widely recognized before.

Abstract

The prevalence of lung cancer as one of the most common cancers with the highest mortality rate is one of the most important health problems in humans across the world. Molecular research can provide valuable information about genetic changes associated with the pathogenesis of the disease that may be used to improve prognosis and treatment. The current study aimed to examine the genotyping utility of the Il12p40 (IL-12B) gene (rs3212227, A>C) polymorphisms and detect its relationship with the concentration levels of HGH and IGF-1 for the non-small cell lung carcinoma (NSCLC). This study investigated 67 cases with NSCLC (60 males and 7 females) and 28 healthy individuals as controls. The serum level of HGH and IGF-1 was determined using an enzyme-linked immunosorbent assay. Genotyping of the IL-12B gene polymorphisms (rs3212227, A>C) was carried out by polymerase chain reaction-restriction fragment length polymorphism. The serum levels of the HGH and IGF-1 were estimated, and the results of the IL-12B genotyping showed an increased risk of NSCLC. The homozygous wild (AA) genotype of the IL-12 gene showed that the risk of NSCLC was higher than that of the heterozygous (AC) and homozygous genotypes (CC). Moreover, a significant elevation was found in the serum levels of the HGH in the NSCLC patients, compared to the control group. The result showed that the IL- 12 gene polymorphism was implicated in the pathogenesis of the NSCLC and directed several metabolic changes.

Abstract

Background: Polyps are pathological concerning histological changes that develop during colorectal and rectal cancer. The degree of polyp severity is due to their ability to transform the polyp cells from normal to malignant. PIK3CA is a gene that plays an important role in cell cycle regulation. Objective: The study aims to evaluate PIK3CA gene expression in tissues of colorectal polyps. Methods: Two study groups employed in the current study were patients (n = 68) and the control group (n = 50). Immunohistochemical techniques were applied to paraffin-embedded tissues to localize PIK3CA expression. Results: The current study showed a significant difference between the control and patients group in PIK staining results; in the control group, there was no case positive for PIK, while in the patient group, all cases were positive. In the rectum, there was no significant difference between cases given positive and negative. Among histological types, according to tubular adenoma histological type, in the right colon, the cases which gave positive for PIK were significantly lower than negative, while in the left colon, the cases which gave positive were significantly higher than negative. Conclusion: The PIK3CA gene initiates polyp formation and benign tumoral growths, which may subsequently undergo malignant transformation.

im: The cytotoxic effects of graphene oxide nanoparticles (GONPs) using MTT assays, observance of apoptotic markers, and oxidative stress were outlined. Materials & methods: Rat embryonic fibroblasts (REFs) and human epithelial breast cells (HBLs) were used at 250, 500 and 750 μg/ml concentrations. Results: Significant cytotoxic and apoptotic effects were observed. Analyses of CYP2E1 and malondialdehyde concentrations in REF and HBL-100 cell lines after exposing to GONPs confirmed the nanomaterials toxicity. However, the glutathione levels in REF and HBL-100 cell lines showed a substantial reduction compared with the control. The cytochrome CYP2E1, glutathione, malondialdehyde and caspase-3 alterations provided a plausible interlinked relationship. Conclusion: The study confirmed the GONPs cytotoxic effects on REF and HBL-100 cell lines. The outcome suggested caution in wide-spread applications of GONPs, which could have implications for occupational health also. © 2023 Future Medicine Ltd. Abstract

Abstract The most common form of graphene in biological science is graphene oxide (GO), which is also the only graphene family of nanomaterials. that necessarily requires a thorough investigation of their biocompatibility as well as a thorough understanding of their cellular interactions by in vivo study. By using Sixty adult male mice which divided into five groups (control,40 mg/kg, 80mg/kg, 120 mg/kg and 200mg/kg) Except the control group, other groups were given graphene oxide nanoparticles (GONPs) with the mention concentrations. In the current study, toxicity of GONPs was investigate after intraperitoneal injection. The result of current study showed there is mild effect on the animal's body weight as well as different effects on the components of blood, that appear in hematological indices such as Hematocrit (Hct), Hemoglobin (Hgb), white blood Cell (WBC), Red Blood Corpuscles (RBC) and platelets (PLT). We can conclude that there are different harmfully effects form graphene oxide on the animal model and according to that on human, Thus the using of graphene oxide should be limited and used specific documented concentration to reduce its unfavorable effects. © 2022 American Institute of Physics Inc.. Abstract

The most frequent intestinal parasites that cause severe disorders in humans are Giardia lamblia and Entamoeba histolytica, which alter serum concentrations of deferent markers due to virulence factors and pathogenicity. A large number of people with infection are asymptomatic, and they can go for up to a year without showing any signs or symptoms. Additionally, due to prolonged diarrhoea but not acute diarrhoea, these parasites can cause malnutrition, weight loss, growth delay, and possibly low cognitive development. The aim of this study is to look at how giardiasis and amoebiasis affect the levels of certain biomarkers in the blood. © 2022, Russian Open Medical Journal.
